KATHMANDU (AFP) - The two archaeologists had a hunch that the Buddha’s birthplace in southern Nepal held secrets that could transform how the world understood the emergence and spread of Buddhism.
Their pursuit would eventually see them excavate the sacred site of Lumbini as monks prayed nearby, leading to the stunning claim that the Buddha was born in the sixth century BC, two centuries earlier than thought.
Veteran Nepalese archaeologist Kosh Prasad Acharya had carried out excavations in Lumbini before in the early 1990s, when Nepal was still ruled by a king and a Maoist insurgency had yet to kick off.
The project ended in 1996 but Acharya remained unsatisfied with the results.
“My belief was that there was another cultural deposit below, which we had not uncovered,” the 62-year-old told AFP.
He headed back to his government job in the capital Kathmandu and waited to retire, restless to return to Lumbini.
The Buddha’s birthplace was lost and overgrown by jungle before its rediscovery in 1896, when the presence of a third century BC pillar bearing inscriptions allowed historians to identify it as Lumbini.
Since then, it has been designated a UNESCO world heritage site, visited by millions of Buddhists every year, with numbers expected to rise exponentially in the following decades.
Acharya had just retired from his last job, as the director general of the department of archaeology, when UNESCO asked him to co-direct an investigation of Lumbini’s foundations.
The cultural organisation asked Acharya and his longtime collaborator, Robin Coningham, Britain’s leading South Asian archaeologist, to head a team that would examine the site so conservators could develop it for growing numbers of visitors.
“In 2010, our first year there, we were pretty much the handmaidens to the conservators,” Coningham told AFP in a phone interview from his office at Britain’s Durham University, which helped fund the UNESCO project.
“The Eureka moment came in 2011, when we came across a brick temple located below the existing Asokan temple, and below that a sort of void.
“It became clear then that there was much more to this excavation.”
Over the next two years, archaeologists, geophysicists and hired workmen from Nepal and Britain worked on the site, digging in the presence of meditating monks and nuns.
“It was a very moving, very special experience to dig for traces while pilgrims prayed and paid homage,” Acharya said.
They dug for a few weeks each year and sent the samples to laboratories for analysis.
Radiocarbon and optically stimulated luminescence techniques were used to date fragments of charcoal and grains of sand found at the site.
The archaeologists also found holes, apparently meant to secure posts, in the open void below the brick temple.
“The intact holes suggested that whoever had built the brick temple had taken care not to damage the ancient structure below, suggesting the site was always considered holy,” Coningham said.
Lab tests confirmed the existence of roots within the void below the brick structure, suggesting it may have been a shrine where a tree once grew, possibly the hardwood sal tree under which many believe the Buddha was born.
The discovery, revealed in November, sparked huge excitement, but some historians have urged caution, saying the ancient tree shrine could have been built by pre-Buddhist believers.
“The worship of trees, often at simple altars, was a ubiquitous feature of ancient Indian religions,” Julia Shaw, a lecturer in South Asian archaeology at University College London told National Geographic’s online edition.
“It is also possible that what is being described represents an older tree shrine quite disconnected from the worship of the historical Buddha,” Shaw added.
According to Coningham, however, if the Buddhists had appropriated the tree shrine from non-Buddhists, the site would not have survived relatively unscathed.
“Also, the inscriptions at Bodhgaya (where the Buddha achieved enlightenment) reveal a thriving culture of tree worship, which suggests continuity,” he added.
Much of what is known about the Buddha’s life has its origins in oral tradition. The earliest decipherable written records in the region, the inscriptions of India’s Buddhist emperor Asoka, are dated about 250 BC.
Prior to this discovery, most scholars said that the Buddha – who renounced material wealth to embrace and preach a life of non-attachment – lived during the fourth century BC, founding a religion that now counts 500 million followers.
Buddhists in Nepal and Sri Lanka, however, have always believed that the sage was born around 623 BC, a date that now seems more accurate.
“It’s one of the great puzzles, this discovery reveals the endurance of oral traditions,” Coningham said.
“This is one of those very rare times when tradition, belief and archaeology all come together.”

Cancer is one of the worst and most common diseases; it affects nearly 1 in 3 people at some stage in their life. For those who say there is not any “credible scientific research” on this fruit: Do you know how much a “credible scientific evidence” costs? Probably tens of millions of dollars, and who might pay for that? NIH? NSF? Pharmaceutical companies? Pharmaceuticals lobby against anything that damages their profit, and NIH and NSF don’t care about “natural alternatives”. The profits that each individual patient brings for pharmaceutical industry is more than a million dollars and they never want to lose that profit. So there might never be a “credible scientific research” for this fruit or any other natural remedies in near future!

Abstract 
Pancreatic tumors are resistant to conventional chemotherapies. The present study was aimed at evaluating the potential of a novel plant-derived product as a therapeutic agent for pancreatic cancer (PC). The effects of an extract from the tropical tree Annona Muricata, commonly known as Graviola, was evaluated for cytotoxicity, cell metabolism, cancer-associated protein/gene expression, tumorigenicity, and metastatic properties of PC cells. Our experiments revealed that Graviola induced necrosis of PC cells by inhibiting cellular metabolism. The expression of molecules related to hypoxia and glycolysis in PC cells (i.e. HIF-1α, NF-κB, GLUT1, GLUT4, HKII, and LDHA) were downregulated in the presence of the extract. In vitro functional assays further confirmed the inhibition of tumorigenic properties of PC cells. Overall, the compounds that are naturally present in a Graviola extract inhibited multiple signaling pathways that regulate metabolism, cell cycle, survival, and metastatic properties in PC cells. Collectively, alterations in these parameters led to a decrease in tumorigenicity and metastasis of orthotopically implanted pancreatic tumors, indicating promising characteristics of the natural product against this lethal disease.

Abstract 
Annona muricata L (Annonaceae), commonly known as soursop has a long, rich history in herbal medicine with a lengthy recorded indigenous use. It had also been found to be a promising new anti-tumor agent in numerous in vitro studies. The present investigation concerns chemopreventive effects in a two-stage model of skin papillomagenesis. Chemopreventive effects of an ethanolic extract of A. muricata leaves (AMLE) was evaluated in 6-7 week old ICR mice given a single topical application of 7,12-dimethylbenza(α)anthracene (DMBA 100 μg/100 μl acetone) and promotion by repeated application of croton oil (1% in acetone/ twice a week) for 10 weeks. Morphological tumor incidence, burden and volume were measured, with histological evaluation of skin tissue. Topical application of AMLE at 30, 100 and 300 mg/kg significantly reduced DMBA/croton oil induced mice skin papillomagenesis in (i) peri-initiation protocol (AMLE from 7 days prior to 7 days after DMBA), (ii) promotion protocol (AMLE 30 minutes after croton oil), or (iii) both peri-initiation and promotion protocol (AMLE 7 days prior to 7 day after DMBA and AMLE 30 minutes after croton oil throughout the experimental period), in a dose dependent manner (p<0.05) as compared to carcinogen-treated control. Furthermore, the average latent period was significantly increased in the AMLE-treated group. Interestingly, At 100 and 300 mg/ kg, AMLE completely inhibited the tumor development in all stages. Histopathological study revealed that tumor growth from the AMLE-treated groups showed only slight hyperplasia and absence of keratin pearls and rete ridges. The results, thus suggest that the A.muricata leaves extract was able to suppress tumor initiation as well as tumor promotion even at lower dosage.

AbstractThe cell inhibition activities of several Annonaceous acetogenins, covering the three major structural classes of bis-adjacent, bis-non-adjacent, and single tetrahydrofuran (THF) ring compounds and their respective ketolactone rearrangement products, were tested in an in vitro disk diffusion assay against three murine (P388, PO3, and M17/Adr) and two human (H8 and H125) cancerous cell lines as well as a non-cancerous immortalized rat GI epithelial cell line (I18). The results demonstrate a dose-dependent inhibition of cancerous cell growth, while non-cancerous cell growth is not inhibited by the same dosages. All of the acetogenins, irrespective of their various structural types, inhibit the growth of adriamycin resistant tumor cells and non-resistant tumor cells at the same levels of potency. These results show that the Annonaceous acetogenins are an extremely potent class of compounds, and their inhibition of cell growth can be selective for cancerous cells and also effective for drug resistant cancer cells, while exhibiting only minimal toxicity to ‘normal’ non-cancerous cells.

AbstractAntinociceptive and anti-inflammatory activities of the ethanol extract from Annona muricata L. leaves were investigated in animal models. The extract delivered per oral route (p.o.) reduced the number of abdominal contortions by 14.42% (at a dose of 200 mg/kg) and 41.41% (400 mg/kg). Doses of 200 and 400 mg/kg (p.o) inhibited both phases of the time paw licking: first phase (23.67% and 45.02%) and the second phase (30.09% and 50.02%), respectively. The extract (p.o.) increased the reaction time on a hot plate at doses of 200 (30.77% and 37.04%) and 400 mg/kg (82.61% and 96.30%) after 60 and 90 minutes of treatment, respectively. The paw edema was reduced by the ethanol extract (p.o.) at doses of 200 (23.16% and 29.33%) and 400 mg/kg (29.50% and 37.33%) after 3 to 4 h of application of carrageenan, respectively. Doses of 200 and 400 mg/kg (p.o.), administered 4 h before the carrageenan injection, reduced the exudate volume (29.25 and 45.74%) and leukocyte migration (18.19 and 27.95%) significantly. These results suggest that A. muricata can be an active source of substances with antinociceptive and anti-inflammatory activities.

Abstract 
Annonaceous acetogenins are a group of potential anti-neoplastic agents isolated from Annonaceae plants. In this study, we purified annonacin, a cytotoxic mono-tetrahydrofuran acetogenin, from the seeds of Annona reticulata and analyzed its biological effects. Herein, we have shown that annonacin caused significant cell death in various cancer cell lines. T24 bladder cancer cells at the S phase were more vulnerable to the cytotoxicity of annonacin. Furthermore, annonacin activated p21 in a p53-independent manner and arrested T24 cells at the G1 phase. It also induced Bax expression, enhanced caspase-3 activity, and caused apoptotic cell death in T24 cells. In summary, these results suggest that annonacin is potentially a promising anti-cancer compound.

The human liver is an important bodily organ essential for living a healthy life. Its primary function is to breakdown nutrients from food we eat and help with digestion. 1. Skin changes
One visible symptom of liver damage is changes in the color of your skin. If your skin starts becoming paler or yellowish and your nail tips turn yellow your liver is not performing well and requires help. Newly yellow tints in skin are caused by the liver not releasing toxins which leads to bilirubin accumulation beneath the skin.
2. Abdominal changes
The liver is located near the rib cage, when liver health starts deteriorating it causes pain in the lower abdomen and ribs. Ascites can develop if the problem goes untreated.

3. Frequent gassy sensation
If you frequently suffer from a gassy sensation accompanied by stool pressure its an indication that the liver is not able to secrete digestive juices which are essential to break down food. It also means the digestive system is affected by liver problems.
4. Tiredness
When the liver breaks down food it releases energy used to perform daily tasks. Getting tired easily is a sign of lack of energy which can indicate the liver is unable to do its job properly.
5. Urine changes
When our liver starts getting damaged the color of urine changes to darker variants of yellow. It may also become bloody or tar-colored. Some think this change in color is due to dehydration, if you are ingesting sufficient fluids it indicates poor liver health.

9. Fluid retention
Fluid retention in various parts of the body, especially feet, is a sign of weak liver. This condition is known as edema and looks like swelling, it can happen in your ankles, hands, feet, and thighs. If you press a finger gently into a swollen area the imprint will stay a few seconds with edema.
10. Stool changes
Changes in stool can indicate liver problems. If you observe traces of blood in the bathroom, call your doctor. Other signs of liver damage associated with stool may include periods of constipation or irritable bowel syndrome.

With borderline personality disorder, a person often has an insecure sense of who they are. Their self-image, self-identity, or sense of self often rapidly changes. They may view themselves as evil or bad, and sometimes may feel as though they don’t exist at all. An unstable self-image often leads to frequent changes in jobs, friendships, goals and values.
Their relationships are usually in turmoil. They may idealize someone one moment and then abruptly and dramatically shift to fury and hate over perceived slights or even minor misunderstandings. This is because people with borderline personality disorder often have difficulty accepting gray areas — things seem to be either black or white.
